Product application: Tris buffer is not only widely used as a solvent for nucleic acids and proteins, but also has many important uses. Tris was used to grow protein crystals at different pH. The low ionic strength of Tris buffer can be used for the formation of intermediate fibers of lamin in C. elegans. Tris is also one of the main components of protein electrophoresis buffer. In addition, Tris is also an intermediate for the preparation of surfactants, vulcanization accelerators and some drugs. Tris is also used as a titration standard.
Production method:
First dissolve Tris with a small amount of double distilled water (300~500ml), add HCl, then adjust the pH to 7.6 with HCl (1N) or NaOH (1N), and finally add double distilled water to 1000ml. This solution is a stock solution and stored in a refrigerator at 4 ℃.
Note: The PH value of Tris HCl varies with temperature, so it should be measured at room temperature, so the measured results are reliable.

Buffer characteristics: Tris is a weak base, and its pKa is 8.1 at room temperature (25 ℃); According to the buffer theory, the effective buffer range of Tris buffer solution is between pH 7.0 and 9.2.
The pH of Tris alkali aqueous solution is about 10.5. Generally, hydrochloric acid is added to adjust the pH value to the desired value to obtain the buffer solution of this pH value. However, attention should be paid to the influence of temperature on pKa of Tris.
