TCEP is a very effective thiol reductant, which is widely used as a quantitative reductant for disulfide bonds in protein chemistry and proteomics. The reagent has good stability and solubility in aqueous solution. It is also stable in acidic and alkaline solutions. Compared with other thiol reductants such as DTT, TCEP is not only a highly efficient disulfide bond reductant, but also does not need to be removed in the crosslinking reaction of some thiol groups. Therefore, TCEP is often preferred in protein chemistry and proteomics research.

Tris (2-carboxyethyl) phosphine hydrochloric acid (TCEP HCL) is a non-volatile solid. It is a strong reductant. It can be synthesized by hydrolyzing tris (2-Cyanoethyl) phosphonic acid by refluxing HCL aqueous solution. It supports a variety of biological applications, such as in vitro and in vivo reduction of disulfide bonds in various peptides and proteins. TCEP is an important chelating agent suitable for various heavy metal ions, such as Zn (II), CD (II), Pb (II) and Ni (II).

chemical property

Colorless transparent liquid, insoluble in aliphatic hydrocarbon, slightly soluble in water, soluble in alcohol, ketone, ester, ether, benzene, toluene, xylene, etc., can be miscible with chloroform and carbon tetrachloride, and has good compatibility with cellulose acetate, nitrocellulose, cellulose acetate butyrate, ethyl cellulose, polyvinyl chloride, phenolic resin, polyacrylate and other polymers.

Relevant introduction

It is well known that trialkylphosphines have the property of reducing disulfide bonds. These compounds have good stability in aqueous solutions, can selectively reduce disulfide bonds, and do not react with other normal active functional groups in proteins. However, due to their poor water solubility and bad smell, they are limited in application. But TCEP solves these problems.

TCEP can selectively and quantitatively reduce alkyl disulfide compounds with poor water solubility in a wide range of pH values. Usually, the reaction can be completed in less than 5 minutes at room temperature. TCEP has no unpleasant smell and is not afraid of oxidation in the air.

matters needing attention:

This product is not particularly stable in phosphate buffer, especially under neutral or alkaline pH conditions. Therefore, if this product is to be used in PBS buffer, the working solution must be prepared and used.

This product is charged in solution, so it is not suitable for isoelectric focusing experiment.

For your safety and health, please wear experimental clothes and disposable gloves.

This product is suitable for the preparation steps of gel electrophoresis and solid phase metal ion affinity chromatography (IMAC). It is used as a reducing agent to break the disulfide bonds within or between proteins. It is also particularly suitable for the reaction of maleimide coupling cysteine residues. It can prevent the formation of disulfide bonds between cysteine residues and will not be like DTT or β- Me itself is easy to react with maleimide. RNA isolation experiments are also commonly used in the tissue homogenization step.

Tris (2-carboxyethyl) phosphine hydrochloride (TCEP HCL) is a thiol free and odorless compound, which is widely used in biochemistry and molecular biology as a disulfide bond reducing agent for peptides or proteins. With other similar reducing agents such as dithiothreitol (DTT), β- Mercaptoethanol( β- Compared with me), TCEP HCL has many advantages: 1) no odor - it can be used on the test bench without a fume hood; 2) High stability - the stability of the structure of the compound makes it unnecessary to take any preventive measures to avoid oxidation during operation, use and storage. It will not volatilize and can be reacted at room temperature. Stable at room temperature and resistant to air oxidation. Stable in water-soluble buffer, acid and alkali solutions. Does not react with other functional groups in the protein; 3) Wide working range of pH - it is soluble in water-soluble buffer of almost any pH value, and the effective working range of pH is 1.5-9.0. The pH value after being directly dissolved in water is about 2.5; 4) Strong reducibility - for most experiments, 5-50mm TCEP · HCL can provide enough molar force to fully reduce the disulfide bond by acting at room temperature for several minutes; 5) Wide compatibility - it does not contain mercaptan groups. It is not necessary to remove this reducing agent in advance for downstream mercapto modification experiments such as maleimide coupling.

Thermo Scientific pierce premium TCEP HCl is the highest quality formula we provide for this disulfide reducing agent, specially characterized for applications where product integrity and risk mitigation are critical.

Features of superior TCEP HCl:

• TCEP - a thiol free, tasteless and powerful reductant for cracking disulfide bonds

• high quality - properties and purity confirmed by multiple tests, including quantitative NMR

• product integrity - enhanced test and characterization level based on standard level

• batch retention - retain sufficient supply of past batches to ensure future process testing

• change management - change control notification (CCN) service

• consistent production - batch specific production document review

Compared with standard grade products, superior TCEP HCl covers (a) more analytical testing and product characterization, (b) higher batch specific information and quality assurance review, (c) retention of a large number of batch samples and, (d) change control notifications, thus providing clearer and clearer quality and product support. TCEP HCl is a crystalline tris (2-carbonylethyl) phosphate hydrochloride with high purity. It is a thiol free compound that can effectively reduce disulfide bonds of proteins and polypeptides, and is tasteless and stable. TCEP can selectively and completely reduce stable water-soluble alkylated disulfides in a wide pH range.

Advantages of TCEP HCl over other reducing agents:

• tasteless - unlike DTT or BME, TCEP has no smell, so it can be easily reduced on the test bench.

• stable in air - the inherent stability of TCEP groups makes it unnecessary to take any special precautions to avoid oxidation when handling, using or storing TCEP.

• high efficiency - for most applications, 5 to 50 mm TCEP provides sufficient molar volume to effectively reduce peptide or protein disulfide bonds within minutes at room temperature.

• compatible - with TCEP, most applications (e.g., histidine labeled protein purification, maleimide coupling, etc.) do not need to remove the reducing agent in advance.
