
Tris,or tris(hydroxymethyl)aminomethane,or known during medical useas tromethamine or THAM,is an organic compound with the formula(HOCH2)3CNH2. It is extensively used in biochemistry and molecularbiology as a component of buffer solutions such as in TAE and TBEbuffers,especially for solutions of nucleic acids.

The conjugate acid of Tris has a pKa of 8.07 at 25 °C, which impliesthat the buffer has an effective pH range between 7.1 and 9.1(pKa± 1)at room temperature.

Silver-containing single-junction pH electrodes(e.g., silver chlorideelectrodes)are incompatible with tris since an Ag-tris precipitate formswhich clogs the junction. Double-junction electrodes are resistant to thisproblem, and non-silver containing electrodes are immune.
